I'm just going to throw some answers/questions into the ring here...

I would stop your virus scanner.
Set Blood to a lower resolution, not DOSBox.
Could be because Blood is accessing the CD-Rom if any, try using Deamon Tools and an ISO.

Don't know anything more to throw in.

Search the web for ThrottleWatch and run it with all 3 graphs enabled, and logging, while playing Blood.

Then post the log file here as a zipped attachment.
