Can someone provide me instructions or a video how to make this unit work with a sound blaster pro 2.0?
I have a midi cable for in and out and the speaker connections to the sound card . Does the external mpu 401 unit have a joystick port on it as well ? I have a external mt-32 unit.

The MPU-401 isn't designed to interface with a sound card but with a Roland ISA interface card like the MIF-IPC or MIF-IPC A. Of course, they are rare and very expensive when they turn up, so there's a replica for that:

there are several versions of the 401 box, only the older and bigger one can work with a MIF-IPC cards
can the mpu-401 official card only do intelligent mode? Or can I just use my SB AWE 64 Gold?
Perhaps send a picture of your MPU-401 breakout box first so that we know which unit you have.
Another alternative (until you find all the components) is to use SoftMPU.
