I found a motherboard (an ECS P6BX-A+) and it has a BIOS update to support >40GB hard drives. However, the update is only supported on 2MB BIOS ROMs; this same board can have either 1MB or 2MB. Could I simply pop in a different size ROM if I get a 1MB board? If so, what exact chip do I buy?

The one I linked to in the thread should also work in your motherboard, but check the data sheet to be sure. What EPROM chip is a 256K compatible replacement for the 128K Intel P28F001-BXT-120?

Don't use EPROM, its programming works totally different.
The first one (EEPROM) should work. But you can also take any used 2Mbit chip for this which you salvaged from a dead/useless mobo.
