1) Modify dosbox.
1a) Modify the main bios of award, phoenix or AMI that doesn't allow disk access
2) make dosbox bootstrap like it does on boch and VM software
2a) Load VGA bios
2b) Load main bios
3) run as is.

The point is how do I mod the main bios?

If I wanted to play with a BIOS in emulation, i'd use PCem

DOSBox covers a biosless, versatile target system
