Hey, here's my situation:

I've got DOSBOX 0.72 for Mac OS X, I've got a copy of what I believe is the original CD release of King's Quest VI. I'm hoping to play the latter in the former, but I don't even know where to begin - all the tutorials I looked at begin with mounting, but by drive letters - drive letters conveniently missing in the way OS X works.

Any help would be greatly appreciated.

Minimax is right.

Basically, what's "C:" for DOS, is "/Volumes/Macintosh HD" for Mac users.

No, MiniMax is right. Such basic information can easily be found in the readme and the guides, not to mention the search function.
