No, unless your setup isn't correct.
The one is a MIDI daughterboard (which requires a sound card with a Wave Blaster header to connect to), the other one a standalone 8-bit card.

Where are you going with all these questions?
First you enquired about the MT-32, its compatible modules (including the MPU-401) and now you're onto the GM compatible devices.

If a device is GM compatible, it will play General MIDI soundtracks.
