Given that question, I'm guessing the drives are simply not connected correctly. So a few notes:
1. You always start with A:. If you have only one drive, it's A:.
2. A: and B: is determined by the position of the drive on the cable. End-connector is drive A:, middle connector is drive B:.
3. You must set the drive type correctly in the BIOS (DD/HD). This is very important for a 5.25" drive, since they differ in tracks making the seek test fail.
4. Older 5.25" drives can be terminated. On the cable end, the termination must be present, otherwise not.
From what I understand, you can't even get a 3.5" drive work. Make sure you solve this first before messing with the 5.25" drive.
