Can I change the default internal drive letter from Z to C, for example. I have a program, that work only on drive Z.

And which program would that be that requires drive Z to operate? Suspicious.... 😕
From DOS 3.2 (don't know about earlier versions) to DOS 7.1 you only had drive letters from A: to E: (unless explicitly setting LASTDRIVE in config.sys.)
So I really doubt, you're using a legal app.
You had to mount a path as C:!
For example: mount c c:\dosbox
Now all the files in c:\dosbox are on your dosbox-C
