I have a text application and requires a COM port. It works great with DOSBox. The application works best with 50 lines. But I have been unable to find any settings for 50 line mode. Scanning this forum has yielded no results.

I tried aspect=, windowresolution=, and scaler= options without any luck.

Not with stock DOSBox. OpenGL-HQ has the ability to stretch window content in any way you like, and I think the Direct3D patch can do that, too.
