They do work (updated a new Windows 2000 Pro SP4 machine a week ago), but you need the right patches and certificates:
Windows Update Agent 7.4.7600.226
rootsupd_a153023b66d29034420aa227ccc2164cff75229e.exe
Then hit Win+R, type wuauclt.exe /detectnow and wait for the yellow shield to appear in the tray. If it doesn't work, look in %WINDIR%\WindowsUpdate.txt what's wrong.
After you install them use PortUp to archive them for feature use. You can also use PortUp to enable Microsoft Update without the need for IE6. Bear also in mind that you will get slightly different updates depending on the version of core Windows components you have (Internet Explorer, Windows Media Player, .net Framework etc.).
