This should be a simple question, nevertheless I could not find a comprehensive answer:

What is the pinout of the Soundblaster Pro2 (CT1600) PC speaker connector (JP1) and how should it be connected to the motherboard?
My motherbard offers the following pins: Speaker signal, NC, Ground, +5V.
I know I could try it out somehow, but I do not want to damage the card / board.

Aren't you supposed to connect the speaker to the sound card... instead of the card to the MB?

No, the PC Speaker header on the motherboard is an Output, the PC Speaker header on the Sound Blaster is an Input. The motherboard resources generate the PC Speaker sound.

For the sake of avoiding broken links with good information down the line, I'm ripping you off, Great Hierophant. Hope you don't mind.

Sound Blaster:
Pin 1 = +5V
Pin 2 = Speaker

IBM PC:
Pin 1 = Speaker
Pin 2 = None/Key
Pin 3 = Ground (No need to connect)
Pin 4 = +5V

Yes, it’s my fault.
