Does anyone know what options I have to choose in D-Fend to make Dark Seed using the full screensize instead of just a tiny bit in the middle?

You'll have to elaborate on "tiny bit in the middle".

If I recall correctly Darkseed works perfectly with 0.65 out of the box.

Nope. DOSBox 0.65 & 0.70 has problem when speaking to librarian 2nd time in day 1. It locks up with black screen. Still not fixed yet!
