I need to get a 16 bit dos application to run on a new laptop with 64 bit windows 10. Will dosbox do that?

If it's a dos game, dosbox is your ticket.
If it's a dos program, maybe but it's not supported.
If it's a Windows 16bit program, you might be in luck through installing Windows 3.1x in Dosbox.

But you might be better off with a Virtual Machine

It is an HVAC control program that needs access to a serial port com1 or com2
