Its my first time to find a 386 motherboard without barrel battery leaking.
I need some help to indentify this motherboard.

its just a very common layout used by many brands which can't be identified.
i have a similar board in orange with soldered cpu, which is unfortunately defective and wouldn't work with 128kb cache.

There's sometimes some text in the PCB in the back near power connector.

The only way to switch is to use a soldering iron 😀

The jumper setting for cpu shows 50m, 66m and 80m (without the "Hz").Also there no option for cpu without jumpers. The 80m (or 40m?) needs at least the jumper n.9 closed

How should I use the multimeter to check the polarity of the external battery header?

Connect one probe to the chassis (or ground pin on the power connector or shiny part around screw holes) and other to the edgemost pins on the header, if you get a match you've identified the negative pin, other end is the positive.
