I am trying to setup this ASUS PCI-486SP3 board I received, ultimately to use an AMD 5x86 133.

The board came to me with an Intel Overdrive DX2ODPR66 in it, and the CPU jumpers are set similar to an Intel DX2 (looking at the manual), but with one variation. There were a few other jumpers not right on the motherboard - I think I have all in order.

System has 32MB (16 + 16) RAM and a VLB vidcard - nothing else hooked up. I powered on to see if it POSTs - no screen image at all. After power on for 7-9 minutes, one speaker beep.

Wondering how the 486 Overdrive differs from a DX2 66 - if that is the issue, CPU jumpers.

I am reluctant to try the AMD 5x86 133, as, even though this is a rev1.8 board, it has a BIOS dated 1995, and I recall using this same board model in 1996, I had to update BIOS (I still have that floppy here from years past) to get AMD 5x86 support. Or...I did update for some reason. The manual I have (that did not come with this very board, necessarily) does not mention 5x86. So - not sure I can drive the 5x86 right now, but should be able to drive / test with this 486 Overdrive.

Any differences to know of? Thanks.

This doesn't look like a problem with the CPU jumpers. From my experience the board will POST with almost any settings, only some CPU-specific features (like WB cache) would not work if the jumpers are not correctly set. Do you have a different video card (ISA or PCI) and different RAM to try out? Speaking of RAM, are you sure yours is FPM and not EDO? Finally, uploading a good photo of the board might help.
