Day of the Tentacle (DotT) is a highly regarded point-and-click adventure title from 1993. There was a floppy and a CD-ROM version released at that time. The difference, other than the media type, is that the CD had voice (speech) throughout the game, while the floppy version only had speech in a couple of parts. The CD version is also known as the "talkie" version; and both the MIDI music and speech are encoded in files. There is debate over which music is true to the original composition, MT-32 or General MIDI. However, it seems like General MIDI has been slightly favored as the MIDI driver of choice.
Once DotT is installed, then the sound and music configuration is set by running the setup utility. General MIDI music is on port 330 and Soundblaster should be selected for the voice option. For enhanced General MIDI, there are many options available; one is the dosbox-svn build here (direct link). Another advantage of this build is the OpenGL-HQ (OHQ) video mode (Ykhwong's build has both these advantages, too). This high quality video mode is especially suited for full-screen display of this title.
Either DotT version will not properly output voice and special sound effects unless a couple of settings are modified in dosbox.conf:
ems=true
cycles=18000
If voice is not heard, then lower the "cycles" count further.
Also, if using the above dosbox-svn build, then these settings will enable OHQ and enhanced General MIDI coupled with a soundfont file (such as Scc1t2.sf2):

ScummVM is an alternate program to run RotT, but it is unable to directly activate an "easter egg" which provides emulation of a previous title, Maniac Mansion (link). DOSBox has no such limitation and works properly with this "easter egg".
