The S switch will not let you use Roland. You need the R switch for that. If you are wanting to have MT-32 music, you won't have the digital sounds. The MT-32 may have some sound samples in lieu of the SoundBlaster sounds, but may not be what you expect.
ScummVM allows you to use combinations of configurations that are not normally possible, like the DOS General MIDI with the Windows high res dialog images in KQ6. You may be able to combine the Roland MT-32 music with the SoundBlaster digital sounds in MI2. ScummVM has Munt built into it, but you will need the ROM files, which I assume you already have.
BTW, you can use Munt with the official build of DOSBox by installing the driver or using the Munt QT app. One of the Munt developers, sergm, has a 0.74 build of DOSBox that has Munt support without all of the other experimental patches.
